I agree with others that you left off so may terrific actors and actresses that play the best friend/sidekick/secondary role that you could've given attention to, not make another bland list that has everyone people who are fans of the genre all know.
- Judy Greer is fabulous all the way back to Fern in Jawbreaker. She's played the friend in too many movies to name and even played the villainy in 13 Going on 30.
- Kathryn Hahn is hilarious and has been in so many movies that people may not know her name, but they'll know her once they see her. She's adorable and her timing is always perfect.
- Joan Cusack has been around since Sixteen Candles in the background and her resume is longer than my arm. Another beautiful woman who has such great timing and is always hilarious.
I could go on, but hopefully this brings home my point.

what about Hugh Grant in nine months you say that movie with him in it and that was a rom com a very hilarious movie i laugh my but off watching this movie every time I watch it and also what about the rom com also with Hugh Grant did you hear about the Morgans also hilarious as well. if your going to do a list of actors in movies include the ones that were their early work as well as their recent ones dont just skip the early work thats also important

uh Kathrine Heigel was in 10 things I hate about you how could you forget that movie it was a rom -com and she was in her teens not to mention confessions of a shop ahollic wow you just totally skipped those why tjhose were her best back in the day before knock up and the other movies that followed

While I don't disagree Julia Roberts should be on the list, I think Sandra Bullock and Drew Barrymore should rank higher. I don't think of Julia Roberts as someone who is known for rom-coms, even though she's been in some.

As for Julia Roberts, my favorite of her films is Mystic Pizza. You could categorize that as a rom-com, although there is more going on than just ROM or COM.
